To look up a word on your NOOK while reading:

Touch and hold on a word while reading a book or a newspaper. It will highlight the word and bring up the Quick Actions Toolbar.

Tap Look Up to see the definition of the word.
If you prefer, you also can look up the word or term online on Google or Wikipedia.
